Quantitative Evaluation of Cisplatin Uptake in Sensitive and Resistant Individual Cells by Single-Cell ICP-MS (SC-ICP-MS)

One of the most commonly used anti cancer agents is Cisplatin (cis-diamminedichloroplatinum II), however it is well known that buildup of intracellular platinum concentration can inhibit the efficacy of the drug. The research presented here proposes a method for total platinum analysis in individual cells via scICP-MS and allows for a potential to determine the uptake of Pt species in both sensitive or resistant cell lines. The method utilizes an Enya Mist nebulizer operated at 10 µL/min sample flow and calls for an addition of 10% methanol to top up each sample. Small amounts of organic modifier allow for better performance of the nebulizer at such low flows by lowering the viscosity of the solution and allowing less liquid buildup at the tip.
